ABSTRACT

Abstract Occurrence of Ureaplasma diversum (U. diversum) has been associated with repro-ductive failures in cattle and detected in pigs with and without pneumonia. However, its rolein the porcine respiratory disease complex (PRDC) is unclear. A cross-sectional study was con-ducted in abattoirs, inspecting 280 pig lungs from eight herds. All the lungs were inspected,processed and classified according to the histopathological analysis. Moreover, bronchoalveolarlavage (BAL) specimens were collected and processed by PCR for detection of U. diversum andMycoplasma hyopneumoniae (M. hyopneumoniae). Ureaplasma sp.---U. diversum and M. hyop-neumoniae were detected in 17.1% and 29.3% of the analyzed BAL specimens, respectively. Theconcomitant presence of both microorganisms was detected in 12.5% of the inspected lungs.Both agents were found in lungs with and without pneumonia. M. hyopneumoniae was detectedin 31.8% of pig lungs with enzootic pneumonia-like lesions, while Ureaplasma sp.---U. diversumwas detected in 27.5% of lungs with these lesions. This descriptive exploratory study providesinformation for future experimental and field-based studies to better define the pathogenicrole of this organism within the PRDC.


Resumen La presencia de Ureaplasma diversum se ha asociado a fallas reproductivas en el ganado bovino y se ha detectado en cerdos con y sin neumonía. Sin embargo, su participación en el complejo de enfermedades respiratorias porcinas (CERP) no es clara. Se llevó a cabo un estudio transversal en matadero, inspeccionando 280 pulmones de cerdo provenientes de ocho piaras. Todos los pulmones fueron inspeccionados, procesados y clasificados según el análisis histopatológico. También se colectaron muestras de lavado broncoalveolar (LBA) y se procesaron mediante PCR para la detección de U. diversum y Mycoplasma hyopneumoniae. Ureaplasma sp.-U. diversum y M. hyopneumoniae se detectaron en el 17,1% y en el 29,3% de los LBA analizados, respectivamente. La presencia concomitante de ambos microorganismos se detectó en el 12,5% de los pulmones inspeccionados. Ambos agentes se encontraron en pulmones con y sin neumonía. M. hyopneumoniae se detectó en el 31,8% de los pulmones con lesiones compatibles con neumonía enzoótica, mientras que Ureaplasma sp.-U. diversum se detectó en el 27,5% de los pulmones con estas lesiones. Este estudio exploratorio descriptivo proporciona información para futuros estudios experimentales y de campo tendentes a definir mejor el papel patógeno de este organismo dentro del CERP.

ABSTRACT

OBJECTIVE To analyze the compatible stability of commonly used intravenous drugs in the intensive care units (ICU), and to provide a reference for improving medication safety in clinic. METHODS The commonly used intravenous drugs in the ICU of Hebei General Hospital were investigated and confirmed in April 1-30, 2022, and used as keywords to retrieve the relevant literature about compatible stability from PubMed, CNKI, Wanfang Data and other databases, and manually filtered with Micromedex database at the same time. Then, the compatible stability results of the included literature were analyzed descriptively. RESULTS Totally 32 commonly used intravenous drugs and 39 mixed infusion combinations were collected from ICU of this hospital. A total of 40 studies were included, only 2 studies followed all quality requirements; 18 studies validated their methods to guarantee correct reproducibility; 33 studies evaluated physical stability, including precipitate formation and pH changes; 32 studies evaluated chemical compatibility, mainly content/concentration changes. A total of 666 possible two-drug combinations were obtained from the included literature, of which 254 combinations of stability data were available, including 176 were stable, 68 were unstable, and 10 were contradictory. Totally 412 combinations had no stability results. Among two-drug combinations in ICU of this hospital, 42 combinations were stable, 14 combinations were unstable, and 2 combinations were contradictory. CONCLUSIONS The pH, solvent, excipients and preparation concentration are the factors that affect the stability. There are drug combinations with unstable compatibility of commonly used intravenous drugs in ICU of this hospital. The stability study methods are limited, and the stability data cannot meet the actual clinical needs.

ABSTRACT

【Objective】 To investigate the profiles of RhC, c, E, and e antigens and phenotypes in 4 704 inpatients from multiple regions, i. e. Nanning, Guangzhou and Shenzhen, and provide data information for compatibility blood transfusion of Rh blood group. 【Methods】 The Rh blood group antigens were detected by microcolumn gel cards from three manufactures. If the test and the control results are inconsistent, a third-party reagent would be used, and traditional tube method for confirmation if needed. The Chi-square test and Fisher's exact test were used to analyze antigen frequency and Rh phenotypes in each region. 【Results】 Among the 4 704 inpatients, the frequency of C, c, E, and e antigen was e(91.77%)>C(85.64%)>c(49.62%)>E(41.60%), and Rh phenotypes distribution was CCee(49.40%)>CcEe(27.53%)>Ccee(8.16%)>ccEE(7.74%)>ccEe(4.89%)>CCEe(0.96%)>ccee(0.83%)>CcEE(0.47%)>CCEE(0.02%). There were significant differences in Rh blood type distribution among Nanning, Guangzhou and Shenzhen(P<0.05). Differences in Rh phenotype distribution between male and female were noticed in Shenzhen(P< 0.05), but not in Nanning or Guangzhou. 【Conclusion】 The distribution of Rh blood group in Shenzhen, Nanning and Guangzhou were significantly different from each other, therefore regional characteristics should be considered when carrying out Rh-compatible blood transfusion, so as to guarantee the security of transfusion and reduce the incidence of unexpected antibodies.

【Objective】 To establish the donor bank for local region and distinguish the donors based on the past blood donation frequency and blood donation type, so as to improve the application efficiency of platelet bank. 【Methods】 1) According to the donation type and frequency of blood donors, the blood donors who had joined China Marrow Donor Program(CMDP)in our center from 2011 to 2020 were screened and classified. They are classified as reserve donors, prospective donors, and active donors. The donors, who met the selection conditions of active donors, were enrolled from all apheresis donors in 2020 to expand the local platelet bank. 2) In 2020, 739 blood donors who met the conditions of active donors were screened(including donors who had entered CMDP), and their HLA-A/B loci were detected by HLA high-resolution genotyping, and HPA was detected by Q-PCR genotyping. 3) The compatible platelets provided by three types of donors in 2021 were calculated. 【Results】 1) Taiyuan platelet bank, composed of donors with different previous donation experiences, had been constructed, including 739 active donors, 3840 prospective donors and 18 715 reserve donors. The composition ratio of ABO blood groups among three types of donors was found to be similar via chi square test; there were more male than female in three groups. 2) In 2021, the ratio of the average redonation by active donors in the platelet bank to the regular donation by the general donors was 14.4∶3.98. 3) Of the 142 compatible platelets, supplied to PTR patients in 2021, 83.8% of them came from the redonation of active donors after registration in the bank, and 9.9% and 6.3% from prospective donors and reserve donors, respectively. 4) In 2021, 28.1% of the stored pheresis platalets in our center had HLA typing data, and 54.2% of compatible platelets were retrieved from the inventory, which timely met the needs of clinical patients. 【Conclusion】 Integrating resources and distinguishing the activity degree of donors in platelet bank can reduce the cost of bank building and improve the application efficiency of the platelet bank.

ABSTRACT

【Objective】 To explore the clinical application of a universal platelet product for emergency, which was prepared by suspending O-type apheresis platelet concentrate in AB-type fresh frozen plasma, so as to improve the platelet support ability in emergency and special treatment. 【Methods】 A retrospective analysis of 21 hematological patients, which was divided into 3 groups of platelet transfusion schemes: universal type, AB type and the same type, was performed to analyze the differences in PLT, dose, 24 h PPR, 24 h CCI, follow-up platelet transfusion units and interval, and adverse reactions to transfusion. 【Results】 1)There was no significant difference in PLT, MPV, PDW, K, MA and pH between the initial O-type apheresis platelets and the finished universal platelets (P>0.05). The titers of both anti-A and anti-B in the universal platelets were less than 2. 2) Twenty-one patients were transfused with universal platelet for 27 occasions[1~5 occasions per person; 1 (0.4, 1.0) dose per time]. No adverse reactions to transfusion occurred. 3)There was no significant difference in the effective rate of 24 h CCI and 24 h PPR between the three groups after transfusion(P>0.05). The time interval of the first subsequent platelet transfusion between the compatible group and the universal group was longer than that in the AB-type transfusion group, with significant difference(P0.05) among three groups. 【Conclusion】 In emergency, the application of this universal platelet product can make hematological patients get timely and effective treatment. Its accessibility and effectiveness can be elevated and the infusion interval can be prolonged by improving the preparation method and storage conditions. This product is not only expected to improve the platelet support capacity for patients with emergency and also those undergoing hematopoietic stem cell transplantation during the blood type conversion period, but also may be a practical method to alleviate the contradiction between platelet supply and demand.

ABSTRACT

Platelet compatible transfusion can effectively solve the immune mediated platelet transfusion refractoriness (PTR), save platelet resources and improve blood safety. This paper comments and prospects the compatibility modes of HLA, HPA and CD36, HLA antibody titer, antigen immunogenicity and the development of platelet compatible transfusion. The pattern of HLA compatible platelets involves the matching in the alleles, antigens and epitopes levels, respectively, as well as avoidance donor specificity antibody (DSA) method. While setting the mean fluorescence intensity (MFI) threshold of avoidance DSA needs to be explored when using the DSA prediction method. Allele specific HLA antibodies can be found in the patients with PTR. Therefore, the patients and donors should be genotyped for HLA-A, -B loci at high-resolution level in order to avoid allele specific HLA antibodies. The immunogenicity of various antigens or epitopes at HLA-A and -B loci are different. Selecting donor platelets with low antigen expression or low immunogenicity may be a way of HLA compatible platelets. As the probability and type of HPA antibody production are different in the various populations, the approaching of compatibility HPA involves allele matching and avoidance DSA. As to CD36, the compatibility mode mainly refers to avoidance DSA, which means blood donors with CD36 antigen type Ⅰdeficiency are preferentially selected, and then those with CD36 antigen type Ⅱ deficiency. In the future, more attention should be paid to the scale up of database capacity and update of the information construction. The time waiting for compatible platelets transfusion in clinical could be significantly shortened if the requiring and matching are only conducted within the inventory and candidate platelets.

ABSTRACT

OBJECTIVE:To establish UPLC fin gerprint of 32 compatible herb pairs with Polygonum multiflorum as the core , and to conduct multivariate statistical analysis. METHODS :UPLC method was adopted. Using P. multiflorum and single decoction pieces of compatible herb as reference ,UPLC fingerprints of 32 compatible herb pairs of P. multiflorum were drawn. Common peaks were confirmed by relative retention time and UV absorption spectrum. Non-supervised PCA and supervised OPLS-DA were conducted by using SPSS 19.0 software and SIMCA 13.0 software. RESULTS :There were totally 12 common peaks in UPLC fingerprints of 32 compatible herb pairs of P. multiflorum . The results of non-supervised PCA showed that the accumulative variance contribution rate of primary 6 principal components was 84.633%. The results of cluster analysis of PCA comprehensive score showed that single decoction piece of P. multiflorum ,compatible herb pairs of P. multiflorum with Lycium barbarum ,Rehmannia glutinosa,Paeonia lactiflora ,Codonopsis pilosula ,Eclipta prostrate ,Angelica sinensis ,Glycyrrhiza uralensis ,Astragalus membranaceus and Ophiopogon japonicus were clustered into one category ;others were clustered into one category. Results of supervised OPLS-DA analysis showed that eigen values of 4 principal components were 2.32,2.61,1.58 and 0.90,respectively. There were differences in the contents of 12 common components in the compatibility of P. multiflorum with tonic medicine and non-tonic medicine. The changes of the content of the components after compatibility with tonic medicine were similar. Common peak 7,4,6,3 were main reasons for the differences (variable importance projection value were all higher than 1). CONCLUSIONS:Established fingerprint is simple in operation ,and can be combined with multivariate statistical analysis to evaluate the content changes of common components of 32 compatible herb pairs with P. multiflorum as the core.

ABSTRACT

The similarity between plant and microbial cells encourage the use of microbial metabolites of halophilic bacteria for the alleviation of salt stress in plants. In the current research work, a compatible solute ectoine extracted from a moderately halophilic bacteria Chromohalobacter salexigens KT989776 was used to enhance flax germination and primary seedling under different levels of salinity. Two successive experiments including germination in Petri plates under six levels of salinity (0, 3, 5, 7, 9 and 11 dS.m-1) and a pot experiment under three irrigating water salinity levels (2, 3 and 4) with two types of ectoine application (spray and soil addition) were conducted. Germination parameters were recorded for the first experiment while a fresh and dry weight of plants and peroxidase activity in addition to sodium-potassium ratio were estimated in the pot experiment. Also, ectoine accumulation in plants was detected using HPLC. Results of LC-MS proved the production of ectoine by C. salexigens KT989776 and ectoine enhanced significantly all germination parameters of flax seeds, decreased sodium accumulation in the plant, increased potassium content, and lowered peroxidase and phenoloxidase activity. Also, HPLC analysis proved that ectoine was detected in all treated samples while not detected in non-treated control.

ABSTRACT

BACKGROUND: The supplementation of betaine, an osmoprotective compatible solute, in the cultivation media has been widely used to protect bacterial cells. To explore the effects of betaine addition on industrial fermentation, Escherichia coli THRD, an L-threonine producer, was used to examine the production of L-threonine with betaine supplementation and the underlying mechanism through which betaine functions was investigated. RESULTS: Betaine supplementation in the medium of E. coli THRD significantly improved L-threonine fermentation parameters. The transcription of zwf and corresponding enzyme activity of glucose-6-phosphate dehydrogenase were significantly promoted by betaine addition, which contributed to an enhanced expression of zwf that provided more nicotinamide adenine dinucleotide phosphate (NADPH) for L-threonine synthesis. In addition, as a result of the betaine addition, the betaine-stimulated expression of enhanced green fluorescent protein (eGFP) under the zwf promoter within a plasmid-based cassette proved to be a transcription-level response of zwf. Finally, the promoter of the phosphoenolpyruvate carboxylase gene ppc in THRD was replaced with that of zwf, while L-threonine fermentation of the new strain was promoted by betaine addition. Conclusions: We reveal a novel mode of betaine that facilitates the microbial production of useful compounds. Betaine supplementation upregulates the expression of zwf and increases the NADPH synthesis, which may be beneficial for the cell growth and thereby promote the production of L-threonine. This finding might be useful for the production of NADPH-dependent amino acids and derivatives in E. coli THRD or other E. coli strains.

ABSTRACT

Objective:To investigate the compatible stability of Xingnaojing injection in combination with 9 common medicines, and to provide a reference for clinical application of this injection. Method:According to the clinical application, Xingnaojing injection was mixed with 9 common medicines and placed in the room under dark and light conditions for 6 h. The appearance of compatible solutions was observed, and the HPLC fingerprint was analyzed by similarity evaluation and principal component analysis(PCA). Result:There were no significant changes in the appearance of compatibility of Xingnaojing injection and 9 common medicines, including piracetam and sodium chloride injection, sodium chloride injection and others. The similarities of fingerprint among compatibility of Xingnaojing injection and 9 common medicines were >0.98 at 0 h of compatibility, 6 h of placement and 6 h of illumination. The results of PCA showed that 9 groups of compatible solutions were clustered into 2 categories, the compatibility of Xingnaojing injection and 8 groups including piracetam and sodium chloride injection clustered into one category, and the relative peak areas of the characteristic components of Xingnaojing injection did not change significantly after compatibility, the compatibility of Xingnaojing injection and Danshen Chuanxiongqin injection clustered into another category, the relative peak areas of some characteristic components of Xingnaojing injection increased after compatibility of 0 h and 6 h,and it was more obvious after 6 h of illumination. Conclusion:The compatibility of Xingnaojing injection and 8 common medicines including piracetam and sodium chloride injection has good stability, while the compatibility has stability problems after Xingnaojing injection mixed with Danshen Chuanxiongqin injection. It is suggested that clinical attention should be paid to their compatibility and rational combination of medicines.

ABSTRACT

The quality control of Chinese materia medica (CMM) is the basis for ensuring the safety and effectiveness of clinical medication of traditional Chinese medicine. The whole process of Chinese medicine processing has a great impact on the final quality. The research and determination of the Q-marker of CMM are of great significance to the substance basis research on CMM, the identification of Chinese medicinal materials, the processing of CMM, and the processing of CMM pharmaceutics. The total quantum statistical moment (TQSM) can fully reflect the chromatographic fingerprints information of CMM, with additive, coupling and strong anti-interference. It can be used for qualitative and quantitative analysis of the whole process of CMM, and can also be used to explore the pharmaceutic rule of Chinese medicine compound and its pharmacokinetic process, which can achieve a comprehensive reflection of the quality of CMM and its compound. Through systematic analysis of the research progress of Chinese medicine Q-marker and the principle and application of TQSM, this paper attempts to provide ideas for the research and determination of Chinese medicine quality markers based on TQSM.

ABSTRACT

Many plants accumulate compatible osmolytes at high levels in plant cells such as amino acids and/or develop special epidermal cell bladders which may serve as external water reservoirs and having small and thick-walled cells in response to water deficit. The objectives of the present investigation were: (i) to study effects of water stress on the anatomical traits and accumulation of free amino acids in quinoa leaves and (ii) to describe differences among drought tolerant and sensitive genotypes in such traits following the imposition of water deficit. A field experiment was carried out in the growing season 2015/2016, using a split plot design with five replications. Main plots were allotted to three irrigation regimes, i.e. well watering (WW) [95% field capacity (FC)], moderate water stress (WS) [65% FC] and severe water stress (SWS) [35% FC] and sub plots to five genotypes. Mean squares due to genotypes, irrigation regimes and their interaction were significant (p≤0.01) for studied leaf free amino acids and anatomical traits. Water stress caused a significant decrease in leaf thickness under WS and SWS, upper and lower epidermis under WS, palisade and spongy layers under SWS, but caused a significant increase in palisade and spongy layers under WS and upper and lower epidermis under SWS. The genotype CICA-17 (tolerant) had the thickest leaf and upper epidermis and second thickest lower epidermis, palisade and spongy layers. Contents of each amino acid were significantly increased due to water stress, except Leucine. Increases in amino acid content increased by increasing severity of water stress. Maximum increase (109.6%) was shown by Threonine under SWS, but minimum (8.08%) was by Arginine under WS. Under SWS, the tolerant genotype CICA-17 showed the highest mean increase percentage (47.9%) in total amount of amino acids relative to WW; it showed the highest increase in all amino acids, especially Proline, Methionine and Phenylalanine.

Objective:To study the stability of safflower yellow for injection respectively mixed with fructose injection,invert sugar injection,invert sugar electrolyte injection,glucose and sodium chloride injection,5% glucose injection,10% glucose injection and 0.9% sodium chloride injection. Methods:The physical appearance,number of particles and pH value were observed at room temper-ature after mixing safflower yellow respectively with above injections for 8 hours at the same concentration as that in clinical use. The content of safflower yellow was determined by high performance liquid chromatography (HPLC). Results: There was no significant difference in appearance,pH or safflower yellow content after the mixing,and the number of insolubility particles met the requirement in Chinese Pharmacopeia (2015 edition).Conclusion:Safflower yellow for injection is stable after dissolved in the 7 varieties of infu-sions in 8h.

AIM To study the influence and action mechanism of effective components in compatible solution of Sparganii Rhizoma-Curcumae Rhizoma on pelvic adhesions of model rats with chronic pelvic inflammatory disease.METHODS Laboratory rats were randomly assigned into six groups,a sham-operated group,a model group,a positive Fukeqianjin tablets group,compatible solution groups (40 g/kg high-dose group,20 g/kg medi um-dose group,and 10 g/kg low-dose group).Except rats of the sham-operated group,those in the other groups were induced into models of chronic pelvic inflammatory disease by mechanical bacterial strain implantation,after which a 20 d corresponding drug administration by oral gavage for each group was initiated.After completion of the last drug administration,Verco criterion was taken as the reference to evaluate pelvic adhesions,HE staining was performed on uterine tissues for pathological scoring,ELASA method was applied to measuring serum IL-1β and TNF-α levels,and Western blot method was used to detect the expression levels of proteins FGF-2 and IGF-1 in uterine tissues.RESULTS Compared with the model group,the high-dose compatible solution group had a much sharper fall in Verco scores (P < 0.05),and remarkably decreased serum IL-1β and TNF-α levels (P < 0.01);both high-dose and medium-dose groups manifested with notably alleviated degenerative necrosis and lowered scores of chronic inflammation infiltration and epithelial hyperproliferation (P < 0.01 or P < 0.05),and a distinct reduction in the expression of proteins FGF-2 and IGF-1 (P < 0.05,P < 0.01).CONCLUSION The significant improvement of the chronic pelvic inflammatory disease in model rats due to the effective components in the compatibile solution of Sparganii Rhizoma-Curcumae Rhizoma may be associated with inhibited release of inflammatory mediators IL-1β and TNF-α,and the reduced expression of proteins FGF-2 and IGF-1 as well.

ABSTRACT

As células-tronco mesenquimais dentais (dMSC) têm sido amplamente utilizadas na engenharia tecidual por sua capacidade de auto-renovação e potencial multilinhagem. Dentre as dMSCs, as células-tronco da polpa do dente decíduo esfoliado (SHEDs) destacam-se pela facilidade de coleta, capacidade proliferativa e tempo sobrevivência. O objetivo do estudo foi avaliar, através de experimentos in vitro, o efeito de diferentes doses do laser de baixa intensidade (LBI) na atividade biológica de SHEDs cultivadas sobre arcabouços de ácido polilático (PLA). Células previamente isoladas e caracterizadas foram cultivadas sobre filmes de PLA e foram irradiadas ou não (controle) com um laser diodo InGaAlP (660 nm, 30 mW, modo de ação contínuo) em um screening de doses (1, 4, 7.5, 15, 22.5 e 30 J/cm²). A proliferação e viabilidade celular foram analisadas nos intervalos de 24, 48 e 72 h, através dos ensaios do Alamar Blue e Live/Dead. A morfologia celular foi avaliada por MEV no intervalo de 72h. A avaliação do estresse oxidativo foi realizada através das dosagens de malondialdeído (MDA) e glutationa (GSH). Densidades de energia de 1 J/cm² e 4 J/cm² promoveram um efeito estimulador da proliferação celular nos três intervalos de tempo quando comparados ao grupo controle e aos demais grupos irradiados. Por outro lado, doses elevadas de irradiação (22.5 e 30 J/cm²) promoveram redução significativa das células. Verificou-se que a viabilidade celular foi afetada ao longo do experimento nos grupos L22.5 e L30, que apresentaram maior quantidade de células mortas no intervalo de 72h. Na análise por MEV evidenciou-se uma maior densidade celular nos grupos L1 e L4, contrapondo-se às SHEDs arranjadas mais dispersamente nos grupos L22.5 e L30. Em relação ao estresse oxidativo, nos grupos L1 e L4 houve elevação dos níveis de GSH e redução do MDA, de modo contrário, nos grupos L22.5 e L30 houve redução dos níveis de GSH e elevação do MDA. Conclui-se que doses baixas de LBI (1 e 4 J/cm²) promovem proliferação e tem efeito citoprotetor em SHEDs cultivadas sobre arcabouços de PLA, enquanto doses elevadas (22.5 e 30J/cm²) tem ação citotóxica e reduzindo a viabilidade e proliferação destas células (AU).


Dental mesenchymal stem cells (dMSCs) have been widely used in tissue engineering for their capacity for self-renewal and multi-lineage potential. Among the dMSCs, the stem cells from human exfoliated deciduous teeth (SHEDs) are highlighted for their ease of collection, proliferative capacity and survival time. The aim of the study was to evaluate, through in vitro experiments, the effect of different doses of low intensity laser (LLLI) on the biological activity of SHEDs cultured on polylactic acid (PLA) scaffolds. Pre-isolated and characterized cells were cultured on PLA films and irradiated or not (control) with an InGaAlP diode laser (660 nm, 30 mW, continuous action mode) in a dose screening (1, 4, 7.5, 15, 22.5 and 30 J/cm²). Cell proliferation and viability were analyzed at 24, 48 and 72 hr using the Alamar Blue and Live/Dead assays. Cell morphology was evaluated by SEM at 72 h. The evaluation of oxidative stress was performed through the dosages of malondialdehyde (MDA) and glutathione (GSH). Energy densities of 1 J/cm² and 4 J/cm² promoted cell proliferation in all intervals when compared to the control group and the other irradiated groups. Contrarily, high doses of irradiation (22.5 and 30 J/cm²) promoted significant negative effect on cell proliferation. It was verified that cell viability was affected throughout the experiment in groups L22.5 and L30, which presented a higher number of dead cells in the interval of 72h. The SEM analysis showed a greater cell density in L1 and L4 groups, opposing the SHEDs arranged more sparsely in L22.5 and L30 groups. Regarding to oxidative stress, in L1 and L4 groups there was elevation of GSH levels and reduction of MDA, showing a cytoprotective effect of these doses. Conversely, in groups L22.5 and L30 there was a reduction of GSH levels and elevation of MDA, corroborating with the results of the aforementioned assays. It is concluded that low doses of LLLI (1 and 4 J/cm²) promote proliferation of SHEDs on PLA scaffolds, while high doses (22.5 and 30 J/cm²) promote cytotoxic and anti-proliferative action in these cells (AU).

ABSTRACT

Objective: To study the compatible stability of cyclophosphamide (CTX) and Mesna admixture for injection in 4 commonly used infusions (5% glucose injection,0.9% sodium chloride injection,fructose injection and xylitol injection) at different temperatures.Methods: An HPLC method was adopted to determine the contents of CTX for injection in 24h respectively under 5℃ and 25℃.The appearance and pH value of the admixture were investigated.Results: No significant changes of appearance, pH value or content of the admixture were found out in 24h under 5℃ and 25℃.Conclusion: CTX and Mesna admixture for injection is compatible with the 4 commonly used infusions under 5℃ and 25℃ for clinical use in 24 h.

ABSTRACT

OBJECTIVE:To investigate the compatible stability of Calcium folinate for injection mixed with Glucose injection and Sodium chloride injection. METHODS:Referring to clinical common concentration,each 3 Calcium folinate for injection (each injection was equal to calcium folinate 100 mg)were respectively mixed with Glucose injection 250 mL or Sodium chloride injection 250 mL. At room temperature,under light or dark condition,the appearance of mixtures,pH value and the number of in-soluble particles were investigated 0,1,2,3,4,6,8,12,24,36,48 h. The contents of calcium folinate in mixtures were deter-mined by HPLC. RESULTS:Under above condition,the color of the mixtures had no change,and no gas,precipitation and turbid-ity was found;there was no evident change in pH values(RSD<2%,n=11). 0 h after mixing,there was large number of parti-cles≥10 μm in mixtures,but the number of particle was decreased as time;within 48 h,the number of particles ≥10 μm and ≥25 μm in mixtures were all in line with the standard of Chinese Pharmacopeia(2015 edition). Under the protection from light con-dition,relative contents of calcium folinate in mixtures had no significant change(RSD<2%,n=11). Under light condition,rela-tive contents of calcium folinate in mixtures decreased significantly,decreasing to 94.5%(mixed with Glucose injection) and 88.4%(mixed with Sodium chloride injection). CONCLUSIONS:Calcium folinate for injection is more stable in Glucose injec-tion,and the stability of compatibility can be affected by light conditions. After mixed with Glucose injection and Sodium chloride injection,Calcium folinate for injection should be kept away from light and used as soon as possible

ABSTRACT

OBJECTIVE:To investigate the compatible stability of Xiao'aiping injection combined with 3 kinds of common in-jections. METHODS:Referring to package inserts,Xiao'aiping injection 40 mL was compatible with 5% Glucose injection,10%Glucose injection or 0.9% Sodium chloride injection 160 mL,respectively. At room temperature(about 25 ℃)and high tempera-ture(40 ℃),the appearance of mixtures were observed at 0,1,2,4,8,12,24,48 h;pH value and the number of insoluble particles were detected. The contents of tenacissoside A and tenacissoside Ⅰ in mixtures were determined by HPLC. RESULTS:Un-der above condition,the mixtures were brownish yellow liquid within 48 h after Xiao'aiping injection was compatible with 5%Glucose injection or 10% Glucose injection;24 h after mixed with 0.9% Sodium chloride injection,the mixture changed from brownish yellow to reddish brown,but no precipitation was found. The pH value of mixtures had no significant change(RSD<1%,n=8). The number of particles ≥25 μm was in line with the requirements of Chinese Pharmacopeia(2015 edition). For-ty-eight hours after mixing,the number of particles ≥10 μm in the mixtures exceeded the pharmacopoeia limits. Within 48 h after mixing,the relative contents of tenacissoside A and tenacissoside I in mixtures had no significant change(RSD<2%,n=8). CON-CLUSIONS:The mixture should be used up within 24 h after Xiao'aiping injection combined with 5% Glucose injection,10%Glucose injection or 0.9% Sodium chloride injection.

ABSTRACT

Objective To compare the clinical efficacy between pediatric ABO-incompatible liver transplantation (ILT) and ABO-compatible liver transplantation (CLT) by Meta-analysis. Methods Relevant studies published until May 2017 were electronically retrieved from PubMed, Embase, Cochrane library, China national knowledge internet (CNKI),Wanfang and VIP databases. According to the inclusion and exclusion criteria, the publications eligible were screened and clinical data were extracted. Meta-analysis was performed using the random or fixed effect model analyses with Review Manager 5.3 statistical software. Results Eleven retrospective cohort studies in English were selected. Meta-analysis demonstrated that the postoperative 1-year survival rate of the recipients in the ILT group was significantly lower than that in the CLT group [odds ratio (OR)=0.64, 95% confidence interval (CI) 0.49-0.83, P=0.0007)]. In the ILT group, the incidence of postoperative rejection reactions was considerably higher compared with that in the CLT group (OR=1.96,95% CI 1.03-3.72, P=0.04). No statistical significance was observed in the postoperative 3- and 10-year survival rate of the recipients, 1-, 3- and 10-year survival rate of the graft and the incidence of postoperative biliary tract complications between two groups (all P>0.05). Conclusions Compared with their CLT counterparts, the 1-year survival rate of the ILT recipients is lower, whereas the incidence of rejection reactions is higher. However, the long-term survival rate of both the recipient and graft and the incidence of biliary tract complications do not significantly differ between CLT and ILT. ILT is a relatively ideal option for emergent patients or those lacking of liver graft with compatible blood group for a long period of time.

ABSTRACT

With the deepening of the study of toxicology of traditional Chinese medicine, the mechanism of hepatotoxicity caused by Tripterygium wilfordii has been gradually revealed. As one of the characteristics of traditional Chinese medicine, Chinese medicine compatibility show its more reasonable and scientific effect in terms of reducing toxicity. In this paper, the author summarizes the research on the basis and mechanism of toxic substances in T. wilfordii, and sum up the compatibility of traditional Chinese medicine (preparation) to reduce its role of hepatotoxicity, so as to provide reference for the rationality and safety in clinical application of T. wilfordii, thereby reducing liver adverse reactions.
